Studi Komparasi Sappanon A dan Sappanon B terhadap Penambatan Protein Tyrosin Phospatase 1B

Abstract


Sappanon A dan sappanon B merupakan senyawa turunan sappanin yang terkandung dalam Caesalpinia sappan  dan menjadi ciri khas dari kandungan metabolit kayu secang. Penelitian ini bertujuan untuk mengidentifikasi potensi aktivitas antidiabetes senyawa sappanon A dan sappanon B melalui penambatan protein tyrosine phosphatase 1B (PTP1B). kajian insilico digunakan dalam penelitian ini yaitu dengan menginteraksikan senyawa sappanon A dan sappanon B dengan protein PTP1B dengan program Molegro virtual docker versi 5. Tahap selanjutnya, hasil docking dianalisis dan divisualisasi dengan PyMol dan Discovery Studio versi 21.1.1. analisis penambatan PTP1B menunjukkan senyawa sappanon A dan sappanon B menghambat PTP1B di daerah yang sama, yaitu daerah inhibitor. Hal ini ditunjukkan dengan beberapa residu yang sama pada sappanon A, sappanon B dan senyawa kontrol inhibitor PTP1B. jenis ikatan pada kompleks senyawa – PTP1B yaitu ikatan hydrogen, interaksi hidrofobik, dan unfavorable bond. Berdasarkan energi ikatan yang dihasilkan, senyawa sappanon A menunjukkan energi ikatan paling rendah yang mengindikasikan ikatan senyawa yang paling kuat. Penelitian ini disimpulkan bahwa sappanon A dan sappanon B berpotensi sebagai antidiabetes dengan menghambat protein PTP1B pada daerah inhibitor.
